 We need to add mandatory comments when do Action that move State to another State

you will have to write a custom advisor to enforce this.. the product does not provide this capability out of the box.
you cannot do it with scripting

Can I in force to add Defect owner after move from State New to In Progress

yes, you can use the Required attributes for type and State advisor
