Basically, I'm trying to add a text field ("Acceptance Criteria") next to the Description field in the Story work item.

I'm trying to modify the layout in order to have 2 columns in one section. I got that by changing the type of the section to "Details" instead of "Description", but the fields end up being too small.

Is there any way to edit these layouts or to create a new one?

No and No.. IBM doesn't provide any edit or authoring mechanism for the layouts.
