In RTC, how can I define a new Work Item Type (other than the existing Defect, Task, Story, Epic, etc.)? Is it possible by some configuration in the UI or is it only through some RTC extension?

You can do that in Project Area configuration editors.
Check Jazz.net articles. There are couple about how to do that step by step.
