We need to fetch the list of users of particular Project area in RTC5 through RTC web client  by constructing workitem query, Please let us know is it feasible or not?

Query is constructed for work item. I don't think you can get the project area member list from a query.
However you can consider to use Reportable API. Please check https://jazz.net/wiki/bin/view/Main/DraftTeamProcessRestApi
